To investigate protein-protein interactions in the actetate biosynthetic pathway, we have developed a suite of tools to modify acyl carrier proteins (ACPs) from fatty acid synthase (FAS) using substrate analogs and reactive probes. These ACP conjugates are used to study the structure of the CPs using NMR and x-ray crystallography. We have developed new tools to capture the dynamic and static interactions between CPs and catalytic partners to understand the mechanisms of substrate binding and protein-protein interactions. Here we highlight investigations of ACP interactions with new FAS enzymes.
