Despite many advancements in the development of delivery vehicles for RNAi therapeutics, none, at this time, have obtained FDA approval. We have designed silica glyconanoparticles in order to investigate key siRNA delivery vehicle criteria including functionalization, size, and surface charge, while maintaining a constant spherical morphology. The Stöber silica nanoparticle synthesis offers a straightforward method of producing biocompatible materials with control of the nanoparticle diameter. This work demonstrates that modified Stöber methods, specifically one that incorporates the polysaccharide dextran, can impart unique particle morphologies that can enhance the siRNA delivery efficiency of silica nanoparticles across multiple cell lines relevant to drug development. This work seeks to delineate the role that dextran plays in the modified Stöber synthesis via physical characterization, as well as in delivery vehicle uptake and gene silencing efficacy.<br/>
