Nickel Acireductone Dioxygenase (Ni-ARD) is involved in the regulation of cellular growth in plants and bacteria. A mammalian analogue has been reported and recently implicated in prostate cancer in rats and human cell lines. At this time only one functional model of the enzyme exists and no structural models of the resting state have been reported. To further understand the influence of the coordination environment on the reactivity of Ni-ARD we have synthesized and characterized the first family of structurally related analogues of the resting state of Ni-ARD. This family greatly expands the available models for the enzyme and aid in enhancing our understanding of how structure influences function. Preliminary reactivity and substrate analogue binding studies will be presented.
