In the present work, electromembrane extraction (EME) and pulsed-electromembrane extraction (PEME) techniques coupled with high-performance liquid chromatography (HPLC) were compared for the extraction of fluoxetine (FLX) and olanzapine (OLZ) from human plasma and urine samples. The influence of extraction parameters affecting the extraction efficiency of FLX and OLZ such as composition of source and receiving phases, extraction time, applied voltage, stirring rate and the duration of pulse and outage periods (in PEME) were studied and optimized.<br/>In PEME, OLZ and FLX were transported under electrokinetic migration conditions in a pulsed electrical field sustained over the SLM (supported liquid membrane). Compared to the EME, in PEME, the system stability is increased by decreasing the thickness of the double layer at the interfaces and extractability is accommodated by eliminating the mass transfer barrier between the two electrodes. According to the results, compared to EME, PEME provides higher preconcentration factors and lower LODs within shorter extraction time from biological samples.
