Host cell impurities remaining after downstream purification of monoclonal antibodies has the potential to illicit adverse immune responses in patients and limit efficacy. Despite an extensive purification process, Chinese hamster ovary host cell proteins (CHO-HCP) have been found to appear with monoclonal antibodies (Mabs) after the purification process regardless of processing conditions. The presence of HCP over a long range of purification conditions suggests the possibility of product association in which HCP may directly interact or bind to Mabs. We have isolated model CHO-HCP transcripts from the CHO-K1 cell line and expressed these proteins on the yeast surface for characterization. We have demonstrated that one of these transcripts, lipoprotein lipase, makes a direct interaction with a collection of human IgG by fluorescence microscopy.  The nature of these HCP-antibody interactions in a variety of solution conditions is currently being examined with the goal of identifying potential wash conditions to eliminate particular HCPs from Mab products.<br/>
