Oxidative DNA damage results in formation of the 8-oxoguanine (8oxoG) lesion, a mutagenic base alteration responsible for G→T conversion. Human 8-oxoguanine DNA glycosylase (hOGG1) is a vital enzyme in the base excision repair pathway and is responsible for excision of 8oxoG from duplex DNA. The activity of hOGG1 has been previously shown to be compromised by common polymorphisms in hOGG1 and to be stimulated by AP endonuclease, the enzyme downstream from hOGG1 in the base excision repair pathway. Here we report our results studying the differential stimulation of hOGG1 mutants by AP endonuclease. Using kinetic and binding assays, we have investigated the extent to which the reductions in hOGG1 activity caused by mutations in hOGG1 can be mitigated by the action of AP endonuclease.
