[HisProGlyGly + H]+ ions were formed in the gas phase by electrospray ionization and analyzed by ion mobility spectrometry-mass spectrometry. Two major conformers were observed, corresponding to cis and trans configurations of the proline residue. These were individually selected and activated over a range of acceleration voltages to measure thresholds for interconversion of the proline configuration. Energy threshold modeling methods were tested on analogous data for a system with known energy barriers, triply protonated bradykinin, and then used to determine the energy barrier to interconversion in [HisProGlyGly + H]+ ions.
