Daniel I.C. Wang developed and implemented ground-breaking technological innovations to enhance mammalian cell cultures. These advances demonstrated the potential of bioprocessing to improve the quality and quantity of biologics production, paving the way for the current biopharmaceutical industry and biomanufacturing research in academia.  It is upon this foundation that many biochemical engineers have built their current research programs.   One of the many research thrusts of Professor Wang is in the area of glycosylation, in which proteins undergo complex post-translational modifications that can alter the structure, function, and in vivo properties of recombinant therapeutic proteins.  A number of factors can impact glycosylation processing in animal cell cultures, and the capacity to both elucidate and manipulate these modifications will be important for biotechnology.   In our studies, we are implementing analytic, computational, and metabolic engineering methodologies to both determine the glycosylation structures attached to heterologous proteins and if desired change these glycoforms.  First, it is important to understand the glycan structures emerging from cell cultures using advanced analytical methods.  Next we may want to modify these structures through changes in the culture environment or modifications to the producer cells. One approach is to alter the media such that the glycosylation patterns change.  Another approach is to engineer the cell lines in order to manipulate the glycosylation capabilities of animal cells.   In concert, mathematical models can be used to evaluate the impact of these changes in order to optimize glycosylation processing and obtain a desired glycan profile.   Together, a comprehensive systems approach will enhance our ability to control mammalian cultures at the cellular and bioreactor levels, in ways that are building on the approaches pioneered by Dr. Wang.<br/> <br/>
