Understanding and determining the ideal cell culture conditions is an important step in the biomanufacturing process. Seeking for optimal cell media and supplement is certainly critical to final production yield and product quality. The initial experimentation with cells cultured in different types of commercially available media in small scale bioreactors facilitates process development and culture optimization. Antifoam supplement studies have become important as excessive foaming in the bioreactors can lead to culture contamination and suboptimal culturing volume resulting in low productivity. In this project, we aim to utilize an AMBR high-throughput micro-bioreactor system to screen for ideal CHO cell growth media and antifoam supplement. With the disposable bioreactors controlled by an automated workstation, a series of experiments were performed to optimize the cell growth. Viable cell density, cell viability, foam height and nutrient concentration were recorded and monitored during the cell culture. The harvested IgG concentration was measured using the Octet systems from each culture condition. Optimal culture conditions were selected based on viable cell density, cell viability and specific productivity. Our study demonstrates the use of the AMBR high-throughput micro-bioreactor system to facilitate the development and refinement of the process involved in culturing CHO cells.
