The rational design of new and improved glycosylation reactions required for the synthesis of biologically relevant glycosides and their conjugates is best conducted on the basis of a sound understanding of the glycosylation reaction mechanism and the factors that influence it. Taking Winstein's ion pair theory as a starting point the lecture will cover ongoing investigations in the Crich laboratory on the glycosylation reaction mechanism and their application in the desin of improved methods. Areas to be covered include i) the design of simple methods for assessing relative reaction kinetics of glycosylation reactions, and ii) the influence of substituents with particular emphasis on side chain conformation.
