We are investigating a nanoparticle therapeutic silica-based (protocell) delivery system to overcome intracellular bacterial infections using bacteriophage. To aid in vivo visualization and tracking of bacteriophage delivery, we are employing fluorescent and electron-dense bioimaging nanoparticles. Bacteriophage are being used as templates to attach commercial and synthesized Quantum Dots (QDs) for future identification of bacteria within cells and tissue. The protein coat of MS2 phage, with exterior –NH2 groups from lysine residues, provides an ideal target for functionalization and conjugation of nanoparticles. QDs were selected due to their tunable emissions properties within the biological tissue optical window and their high quantum yields. Several conjugation strategies are being explored including EDC/NHS coupling to attach the QDs to the MS2 protein coat. Details on the results of viability studies using plaque assays and silica encapsulation will be presented.
