Deoxypodophyllotoxin (DPT) is a natural product extracted from Anthriscus sylvestris that has potent anticancer and antitumor activity (IC50 13-25nM). F4-4 a related unpublished compound isolated by Dr. Rex Cates (BYU Biology) from A. silvestris has potent antiviral activity against Herpes simplex viruses 1 (HSV-1) and 2 (HSV-2) and Herpes zostera (&gt;5mM). Both F4 and DPT are scarce in natural sources, therefore synthesis is required. We present here the progress toward the total synthesis of DPT and F4-4 using the key styryl-alkyne IMDA reaction that to our knowledge has not been used in a total synthesis of natural products. We have completed the racemic synthesis of DPT.
