The intricate roles RNA play within cellular function beyond serving as translational templates are of immense and rapidly growing interest, but discovery has been hindered in part due to a limited RNA-centric toolbox. Bioorthogonal approaches to visualize and capture RNA to discover RNA’s non-coding functions are rapidly being developed to fill this need. We recently demonstrated both antisense targeting and covalent labeling approaches to monitor and track specific RNAs in fixed and living tissues. We have continued to develop both our covalent labeling strategy, RNA-TAG (transglycosylation at guanine), and RNA hybridization based labeling approaches that harness tetrazine mediated uncaging reactions to investigate RNA biology. Here we provide an update to our current approaches to detect, image, and modify specific RNAs of interest and demonstrate applications of these technologies to discovery of noncoding RNA function.
