Stem cell shows great potential for curing many diseases, but its efficacy is low due to the injection to fibrotic tissues, low cell retention at the diseased area, and poor survivability after transplantation. Here, we tried to overcome these limitations by engineering cells with an integrated nanoparticle (NP) namede as mesocellular silica-superparamagnetic iron oxide (MCS-SPIO). This survivability of human mesenchymal stem cells (hMSCs) incubated with 1 mg/ml NP was 99%.Proliferation assay showed the doubling time for labeled and unlabeled hMSCs were 2.31 and 2.27 days. The NP exhibited a high loading capacity of IGF 2.28 µg/mg NP, more than 4.56-fold higher than previous report. The IGF loaded NP (IGF@MCS-SPIO) shows a burst release in the first 10 hours and a continuous release for up to one week (Fig. 1A).The IGF@MCS-SPIO increased the hMSCs survivability after one week compared to free IGF (Fig. 1B ). The NP can enter hMSCs (Fig. 1C) and increase their ultrasound signal (Fig. 1D). ICP-OES experiment showed there were 0.038 ng Si and 0.035 ng Fe per cell. Moreover, in vitro experiments showed the combination of NP and magnetic field can make the cells grow on the side of flask (Fig. 1E), which is promising in increasing the in vivo retention. The calcein/EthD-III assay and flow cytometry results indicated most cells were alive after treated with MCS-SPIO and magnetic field (Fig. 1F). A cell migration assay demonstrated these treated hMSCs migrated to an area depleted of hMSCs within 72 hours (Fig. 1G). Future work will utilize these materials with small animal models of human stem cell therapy to measure the impact of magnetic retention on ejection fraction.
