Reactive oxygen species are a series of highly reactive molecules that mainly found in the redox reaction organelles. ROS is produced from the organelles using oxygen. such as mitochondria and endoplasmic reticulum etc.. ROS draws more and more attentions of biologist and chemists not only for its damage to DNA but also the role as second messenger in the cell. High concentration ROS can induce the apoptosis of cells. Hydroxyl radical is especially bad since it reacts indiscriminately with biomolecules. Nowadays, many antioxidants have no selectivity to the cells. They will induce the apoptosis of not only the damaged cell but also the normal cells. Our lab found out a nontoxic self-cyclization scaffold can specifically release the drug into only the damaged cells. So l am designing novel antioxidants can selectively response to the hydroxyl radical to recover the damaged cells base on what we found. Two molecules, called AP1 and AP2, were synthesized by six steps of each. Besides showing the synthesis steps and characterization of it by NMR and Mass spectroscopy, I examined the reactivity of them both with HPLC and found that AP2 has better selectivity to be oxidized by hydroxyl radical. The mechanism of the release of drug under hydroxyl radical environment( other ROS after that) will be proved by further biochemical methods. The cytotoxicity test can reveal the ability of them to protect cell from ROS. More biological tests need to be done and they will prove the biochemical potential of AP2.
