Lipid based materials have demonstrated recent success in nanotherapeutics due to their biocompatibility and dual hydrophilic/lipophilic loading capacity. However, trouble still lies in the lack of <i>in vivo </i>stability and controlled guest release for lipid based nanomaterials. This can be solved through the use of polymer nanotherapeutics, which have higher stability and controlled release mechanisms. One caveat to polymer nanomaterials, however, is their inability to non-covalently entrap hydrophilic guest molecules in aqueous milieu. In this work we describe the formation of hybrid lipid coated polymer nanogels (lipogels), which form a core-shell structure capable of dual guest loading with enhanced stability and controlled release, a feat that cannot be achieved with lipid or nanogel systems individually. Herein a lipogel scaffold has been established through a convenient one-pot method of formation, where self-assembling liposomes act as a template for the polymer nanogel core, which is then covalently crosslinked under UV irradiation. The simplicity of this complete aqueous formation allows for easily tuning core-shell characteristics with the capability to encapsulate sensitive biomolecule guests. Such tunability has been displayed through forming a series of lipogels with core and/or shell stimuli-sensitivity (redox and/or pH, respectively), capable of dual guest loading (proteins and/or hydrophobic small molecules). Characterization of lipogel formation, guest encapsulation, stability, and stimuli-sensitivity will be discussed for this particular pH-redox sensitive lipogel series. Additionally, minimal toxicity and intracellular delivery capabilities for these lipogels from <i>in vitro</i> analysis demonstrates the potential for lipogels as a viable nanovehicle for biologic delivery.
