The spectroscopic inaccessibility of the d10 Zn(II) ion is often subverted by its substitution with the d7 Co(II) ion. However, while the Co(II)-substituted enzyme typically gives rise to easily detectable signals in optical and magnetic resonance spectroscopies, interpretation of the data in terms of structure is often difficult due to complicated electronic structure. Nowhere is this more evident than in EPR studies of Co(II)-substituted enzymes. Detailed studies of several series of structural models will be presented that shed light on the notorious complexity of high-spin Co(II) EPR and offer insight into the historical difficulty and future development of more sophisticated interrogations, such as ENDOR and ESEEM.
