Macrocyclization has been a common approach to enhance conformational stability of molecules designed for recognition by proteins, including therapeutic targets. Access to a large diversity of differently functionalized macrocyclic scaffolds will enable their exploration as bioactive molecules. To generate a diverse set of compounds, we have developed a method for the solid-phase synthesis of macrocyclic aromatic oligoamides. Applying this method allows access to analogs that vary ring size, ring flexibility, and appended functionality. This robust and flexible method will expand the range of macrocycles available for future application.<br/>
