In the last three decades, metabolomics has come to the forefront and contributed significantly to drug discovery, biomarker identification and integration of the “omics” sciences. The identification of metabolites from complex metabolomics samples is key to understanding a biological systems response to stress (e.g., disease, drug treatment, etc.); therefore, the goal of this study was to expand the coverage of the metabolome and reveal more pathways by creating an optimized growth media. Using metabolite extracts from previous cell cultures and supplemental commercial extracts (Isogro®), various nutrient rich 13C-labeled media. E. Coli cell cultures were created using 13C-labeled media and the metabolome was extracted for analysis. The E. coli metabolome extracts were compared and analyzed with 2D 1H-13C heteronuclear single quantum coherence (HSQC) NMR experiments and multivariate statistical techniques. An optimized growth media would increase the relative sensitivity (increase biomass) and the number of detectable 13C-labeld metabolites compared to the traditional approach of minimal media supplemented with 13C-glucose. Preliminary results indicate that a culture media consisting of a 0.2% carbon source, 0.2% 13C-labeled metabolite extract, and 3 mg/L of ferrous sulfate significantly increased the concentrations of 13C-labeled metabolites in metabolomics samples. Interestingly, the Isogro® supplement yielded a different set of observable metabolites while also increasing relative metabolite concentrations.
