<br/>This presentations gives a detailed description of when and how discretized multi-level elution (DiME) trajectories can be used to improve separation and process performance for preparative chromatography. Equilibrium theory is used to give an intuitive motivation to when general gradients can be applied and how they will affect the separation. From this description a dynamic optimization problem can be formed, which will govern the shape of the gradient. To solve the optimization problem a model, which can predict the separation on the column, is needed.<br/><br/>To show the applicability of DiME trajectories two case studies are presented. The first case study is a ternary protein separation by batch chromatography. The separation has been optimized and the optimal elution scheme has been experimentally validated on an ÄKTA Pure. The second case study is multi-column counter-current solvent gradient purification (MSCGP) of the same ternary protein mixture. Since MCSGP is a semi-continuous multi-column process the elution scheme is more complex and the elution profiles needs to span over two columns. By using the same methodology as for batch chromatography the optimal elution gradients can be found for the MCSGP case.
