Near-infrared (NIR) spectroscopy has been proposed to be used in the biopharmaceutical industry as a way to monitor cell culture quality in real time, and has been supported by lab scale R&D studies.  In this case study, we have integrated a Sartorius BioPAT ® Spectro NIR in to 5L glass benchtop bioreactors, using a unique flow cell technology.  Preliminary bioreactor runs using CHO cells producing a model IgG1 were used to optimize subsequent bioreactor runs used to build NIR models. Using data collected over multiple bioreactor runs, Partial Least Squares regression models were built to monitor and predict values for glucose, glutamine, glutamate, and lactate.  This unique flow cell allows for additional probes to be installed in to the bioreactors during the culture, allowing for orthogonal measurement of culture nutrients and metabolites by traditional analyses.  Model building was done using both Sartorius SX-Suite/Plus software and SIMCA Online.<br/>
