Nanomedicine has become one of the most important strategies to minimize the lack of drug solubility and drug inactivation after administration into patients. Lack of selectivity is one of the biggest problems to overcome in the development of effective drug delivery systems. Consequently solid-lipid nanoparticles (SLN) have been studied due to their potential simultaneously increase the drug solubility and biological targetability for cancer therapy. To accomplish targeted SLN, transferrin (Tf) and Hyaluronic acid (HA) were incorporated in our system due to the overexpression of their corresponding receptors CD71 and CD44 in many tumors. Herein we developed a SLN (~200 nm in diameter) formulation by the use microemulsion method using stigmasterol (Stg) as the cytotoxic agent. Our system demonstrated strong cytotoxicity on lung cancer (A549 cell line) with an LC<sub>50</sub> of 11 µM after 24 h and caspase-independent apoptotic pathways induction. More in vitro studies will be completed to confirm the targetability of the Stg-SLN with HA-Tf modification and also to elucidate the apoptotic pathway.<br/>This research was funded by the NIH Research Initiative for Scientific Enhancement (RISE) Program Grant No. 5R25GM061151-15
