Stable free radicals are important probes for biological systems; not only can the unpaired electron be detected directly in the diamagnetic background, but it also can quench local fluorophores and influence relaxation times of nearby nuclei. Though as yet little used, verdazyls are particularly interesting in this area because of their stability; unlike nitroxides they are not reduced by aspartate; neither do they react with hydrogen peroxide. We recently reported a series of verdazyl substituted sugars. Here we report our attempts to develop a verdazyl substituted amino acid that can be incorporated into peptides to provide a structural probe.
