Polysulfonated polymers have recently attracted a lot of attention for various biological applications, this is due to their structural similarity with heparin. Here, we have studied the use of reversible addition-fragmentation chain-transfer (RAFT) polymerization to control the polymerization of 2-acrylamido-2-methylpropane sulfonic acid (AMPS) in aqueous solution, and obtained a range of well-defined homo- and block-polymers with various architectures. Excellent control over the polymerization enabled the synthesis of high-order multiblock polymers, as well as core cross-linked star copolymers with alternating AMPS and <i>N</i>-hydroxyethyl acrylamide (HEAm). The resulting materials have been evaluated for their ability to stabilize basic Fibroblast Growth Factor (bFGF), and promote the growth of fibroblast cells.
