Toll-like receptors (TLRs) have been shown to be vital in immune activation. Previously, we have shown control of immune cell activation with photocaged agonists in vitro. Here we present photo control of cell migration in vivo. By strategically protecting the TLR2 agonist, Pam2CSK4, with photocleavable NPPOC, TLR activity is knocked out. NPPOC-Pam2CSK4 can be used to label cells ex vivo and the abeled cells can be adoptively transferred into the footpad of a mouse. We found increased cell migration to the draining popliteal lymph node on mice after light exposure. RNA analysis of the popliteal lymph nodes also showed increase in inflammatory cytokines following light exposure.
