The Enhanced Permeability and Retention (EPR) effect, an idea that focuses on the differences between normal tissues and cancer tissues, has served as a valuable strategy in cancer drug design. Due to the “leaky” nature and flawed drainage system of cancer tissues, macromolecules can enter tumor cells and accumulate, compared to the very effective lymphatic system of normal human tissues. Through this discovery, tumor cells can be targeted through the use of biocompatible macromolecules bearing chemotherapy drugs, thereby minimizing harm to the normal cells surrounding the infected area. Recently, we have begun collaboratively investigating gold nanoparticle aggregate preparation in an effort to make such biocompatible macromolecules.<br/><br/>The purpose of this project is to make linkers that are biocompatible, user friendly, and cost efficient, while retaining the chelation capacity used in past approaches. The ideal linker, the diester of 1,2-dithiolane-3-carboxylic acid linked to triethylene glycol, has been prepared. Discussed herein will be the synthetic route by which the linker was prepared.
