The proliferation of high-throughput cell culture technologies, such as ambr® 15, has allowed upstream development groups to explore a large number of conditions in a short period of time.  Since a number of product quality attributes cannot be measured directly from the cell culture medium, the bottleneck then shifts to clarification and purification of these samples.  An automated liquid handling system with a 96-channel head can be used in conjunction with resin-filled pipette tips.  Purification of 96 samples can be done in as few as three hours, producing about 5 mg of product per sample.  Tips were designed so that the resin bed could fluidize, leaving additional interstitial space between particles compared to a packed column.  Tip design and method parameters were optimized to ensure accuracy and robustness.  The feasibility to utilize the fluidized bed tips for purification of unclarified samples has been demonstrated with no impact on recovery or product quality.  The method was successful even with cell densities above 20 million cells/mL or cell viabilities below 60%.<br/>
