Phenanthridinones are widely found in bioactives. Many derivatives of phenanthridinones have demonstrated 1) anti-HIV activity by inhibiting HIV integrase, 2) cytoprotective and neuroprotective properties, 3) radiotracer properties beneficial for imaging necrosis, 4) the ability to limit the cytotoxicity of doxorubicin, 5) as well as antimicrobial and antifungal abilities (crinasiadine). Herein, we report a novel synthesis of phenanthridinones from N–methoxybenzamides using an oxidative C-H amidation reaction at room temperature with iodobenzene (PhI) catalysis in open air with modest to excellent yields. This method demonstrated an unprecedented substrate scope. In particular, it solved the long-standing challenge in the synthesis of phenanthridinones using sterically demanding substitutions.
