Cells are open systems that are consistently exchanging energy and matter with the environment to maintain homeostasis. As the function is determined by the structure and the structure is determined by the environment, the activity of proteins is sensitive to changes in the environment. Specifically, changes in the temperature, pressure, pH, and ionic strength influence how small molecules bind to proteins. Computer simulation and modeling techniques have been employed to determine how perturbations to the pH and ionic strength alter how protein kinases interact with other proteins and recognize small molecules. Analyses of the relationship between structure-activity and conformational-activity indicate that the protein-protein interactions and the binding of small molecules are sensitive to changes in the pH and ionic strength.
