An unmet need in nanomedicine is to prepare biocompatible and renal clearable nanoparticles by controlling the diameter, composition and surface property of nanoparticles. This paper reports cellulose nanofibers (CNFs) templated synthesis of ultra-small bismuth nanoparticles, and their uses in enhanced X-ray radiation therapy. The interstitial spaces between adjacent fibers are believed to be the adsorption sites of bismuth ions and also to stabilize nanoparticles generated from chemical reduction. The sizes of nanoparticles are controlled in the 1-10 nm range using CNFs with various amount of carboxyl groups. The bismuth nanoparticles show low cytotoxicity alone, and have high yield of reactive oxygen species (ROS) generation in the presence of X-ray radiation. The in vivo animal tests with 4T1 tumor-bearing BALB/C mice show that bismuth nanoparticles can significantly inhibit tumor growths in the presence of X-ray radiation, compared with those with X-ray radiation alone and nanoparticles alone.<br/>
