Hydrogen sulfide (H2S) is now accepted as an important physiological mediator and signaling agent, joining CO and NO as an endogenous gasotransmitter. In its recent emergence as an important biological molecule, investigations have already revealed diverse and important roles of H2S in many aspects of human health and disease. Despite this broad importance, the mechanisms by which H2S exerts its biological action, such as interactions with transition metal centers and the sulfane-sulfur pool, remains underexplored. Motivated by these key challenges, our group is developing purpose-inspired chemical tools for H2S investigations as well as bioinorganic synthetic models to study the action, signaling, and translocation of H2S in biology. This presentation will focus on recent work from our group highlighting insights obtained from model bioorganic and bioinorganic scaffolds lab using transition metal model compounds, synthetic small-molecule isolated persulfides, and synthetic molecules able to bind H2S reversibly through weak interactions. All of these studies highlight the important and expanding roles of H2S in bioinorganic chemistry.
