H2S is a signaling gas that plays important roles in physiological pathways, and exogenous H2S administration provides therapeutic effects in animal models of inflammation, chronic wounds, and others. Although there are many small molecule H2S donors and various methods to deliver H2S, the bottlenecks of many therapeutic H2S delivery studies are rapid release and/or lack of localized delivery. To solve this problem, self-assembling aromatic peptide amphiphiles with attached H2S releasing S-aroylthiooximes (SATOs) were synthesized, and the corresponding hydrogels were produced upon gelation of the aromatic peptide amphiphiles. These aromatic peptide amphiphiles release H2S upon reaction with cysteine, facilitating the localized delivery and slow release of H2S. We studied the self-assembled structure of this family of peptides using a variety of spectroscopic techniques and correlate this with release rates. These hydrogels are a suitable approach for therapeutic gasotransmitter delivery.
