The biodiversity of the marine environment constitute a practically unlimited resource of new multifunctional protein in the field of the development of marine bioactive substances. A Gram-positive, rod-shaped bacterium, designated as Bacillus sp. N6-2, was isolated from surface water from Antarctic sea. A cytotoxicityβ-glucanase from Bacillus sp. N6-2, named NCBG, with a MW of 24.3 kDa was isolated from the fermentation broth of Bacillus sp. N6-2. MTT results showed that NCBG exhibited cytotoxicity against BEL-7402, HepG2, Panc-28, A549, NCI-H460 cells and low cytotoxicity to L-02 cells. Furthermore, Ginhibited the migration and invasion of A549 cells. NCBG was inferred as a kind of β-glucanase by the Mascot search results and N-terminal sequences. According to the result of substrate specificity, NCBG could be proved as a kind of β-glucanase. To our knowledge it is the first time to report the cytotoxicity activity of this protein. NCBG maintains considerable β-glucanse activities and stabilities at wide temperature and pH ranges, and optimum activities are detected at 40 °C and pH 6.0, respectively. Moreover, the specific activity was enhanced by Mn2+. NCBG will more and more widely used in beer and drug industry, health products industry because of its particular cytotoxicity activity and β-glucanase characteristics. Overall, NCBG has the potential to be a multifunctional protein for human health.
