M. tuberculosis is the causative agent of tuberculosis, which is resposible for approximately 2 million deaths each year.The bacteria encodes several metal transport systems that are critical for its survival. KmtR is the second Ni(II)- and Co(II)-responsive metalloregulator in the bacteria. Understanding why M. tuberculosis requires two Ni(II)- and Co(II)-responsive metalloregulators for export, and how they differ, will further our understanding of how the bacteria regulates the concentrations of these essential, but toxic (at high concentrations) metals. Nothing is known about why KmtR is responsive to Ni(II) and Co(II) binding. Our research seeks to determine the metal site structures of KmtR and determine the respective metal binding affinities.
