N-carboxyanhydride (NCA) ring-opening polymerization has enabled the tailored design of polypeptides with low toxicity, a variety of functional groups, and three-dimensional secondary structures due to inter- and intramolecular chain association. We demonstrate new tools in NCA polymerization that increase the utility and function of synthetic polypeptide materials. This includes the synthesis of photoliable amines for the light-induced polymerization of NCAs, providing spatiotemporal control over polypeptide and block-polypeptide synthesis. Additionally, we outline the development of reactive polypeptide scaffolds via the synthesis of controlled molecular weight polysuccinimide and succinimide copolymers from poly(β-L-aspartate).
