Herein, a N-diazeniumdiolate NO donor-modified hyperbranched polyamidoamines (h-PAMAM) is reported with similar molecular weight to third generation PAMAM dendrimers (G3-PAMAM) dendrimers. By changing the extent of propylene oxide (PO) modification, a wide range of NO storage capacities (~1.33-2.50 µmol mg-1) and NO-release kinetics (t1/2 ~34-83 min) were achieved using the h-PAMAM scaffold. Bactericidal action was examined against common dental pathogens including Gram-negative periodontal pathogens (i.e., Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans) and Gram-positive cariogenic bacteria (i.e., Streptococcus mutans and Actinomyces viscosus) as a function of NO release and degree of PO modification. The cytotoxicity of these scaffolds was also evaluated towards human gingival fibroblasts, with results indicating that the combination of NO release and PO modification yields h-PAMAM materials with superior antibacterial action without significant cytotoxicity to mammalian cells. Despite structural defects, the h-PAMAM exhibited comparable biological properties to defect-free G3-PAMAM.
