Two-dimensional separations, especially those done in the liquid phase, are becoming more prevalent due to the availability and evolution of suitable instrumentation. Even in two-dimensions there are still issues about similar compounds not being able to be resolved, especially for complex biomolecule separations. In this talk we explore the feasibility of three-dimensional (3D) separations. 3D separations have been reported in the literature but have not evolved, due to a number limitations which involve sampling speed, orthogonality and methods development. We have recently developed orthogonality metrics that can handle 3D and higher D separations data and we will show where these higher-D separations can be utilized in separations of peptides and glycan analyses.
