Electron-vibration-vibration (EVV) 2DIR is a frequency-resolved spectroscopic technique in which three picosecond laser pulses are overlapped in a sample and the emitted photons are detected whilst the frequencies of the two mid-IR beams are systematically varied, probing vibrational couplings. EVV 2DIR spectra has been measured of the binding domains of plant and mammalian proteins HPPD and FGFR1 respectively, both in complex with and without their corresponding inhibitors NTBC and SU5402. Figure 1 shows HPPD in complex with NTBC, while Figure 2 shows FGFR1 in complex with SU5402. We have shown the ability of this preliminary EVV 2DIR data to detect inhibitor binding as well as the potential to combine experimental and calculated data to extract information about the nature and geometry of interactions.
