Growing market for anti-obesity functional products needs development of high level functionality and fast launch of a new product. Therefore, it is required to develop a novel standard method for evaluation of the anti-obesity functionality.<br/>In numerous studies, the anti-obesity effects have been determined by measurement of changes in white adipose tissue (WAT). The WAT comprises lipid-laden white adipocytes of which new formation and an increase in the size through the differentiation program are essential for WAT remodeling and obesity development. The differentiation program of adipocytes is regulated through the complex interrelationship of several transcription factors (TFs). For example, at the initial stage, stimulation with adipogenic hormones leads to an increase in the activity of CCAAT/enhancer binding proteins β (C/EBPβ). Then, the activity of peroxisome proliferator activated receptor-γ (PPARγ) increases and is maintained at a certain level during the differentiation program. In addition to C/EBPβ and PPARγ, several other TFs have been shown to impact the differentiation program.<br/>In our previous study, we developed a primary frame of the network which consisted of several key TFs (i.e. PPARγ, C/EBPβ, CREB, NFAT, FoxO1, and SREBP-1c) during the adipogenic differentiation. In this study, the network is expanded with additional TFs to evaluate changes of the differentiation program and influence on anti-obesity effects with a broad spectrum. Then, a standard dynamics of the TF network for a positive control (e.g. well known anti-obesity natural products) is determined. Comparison with the standard dynamics of the TF networks for the adipogenic differentiation enables us to test anti-obesity effects of candidate substances quantitatively. The entire process of this study will result in a novel platform of the standard evaluation method for the anti-obesity effects and be applied in many anti-obesity market.
