Since its introduction in the early 1960s, supercritical fluid chromatography (SFC) had enjoyed a rapid rise through the 1990s, before starting a slow decrease in the shadow of other separation techniques. However, a renewed interest in SFC has emerged in the last decade, as the advent of the latest generation of commercial instruments has given new impetus to the development and application of SFC-based methodologies. The application of SFC in pharmaceutical development has been explored at Bristol-Myers Squibb for both chiral and achiral separations. This presentation will demonstrate through several recent case studies the value of SFC as an alternative or orthogonal to conventional chromatographic techniques and its application to a wide range of pharmaceutical sample types and matrices. For example, compared with conventional LC conditions, SFC chiral separations have achieved significant improvements in resolution while shortening the total run time by up to threefold, with the added benefit of significant reduction of organic solvent consumption. Additionally, a comparison of SFC and RPLC for impurity profiling of a fixed dose combination (FDC) drug with a wide polarity range of analytes has been investigated, as well as the application of SFC for some unique, challenging separations that could not be achieved by RPLC.
