Recombinant human growth hormone (rhGH) is use widely in the clinic, especially in pediatric medicine. However, rhGH has a short half-life and the product requires frequent subcutaneous injection leading to poor patient compliance. Currently, it is challenging to develop effective rhGH sustained delivery systems that can exceed a month in duration. In this study, a new sustained release strategy of rhGH was developed by encapsulating sugar glass-stabilized rhGH in electrospun, bioresorbable poly(ester urea)s (PEU) nanofibers. The protein was found to be randomly dispersed throughout the fibers in an aggregate form, and sustained rhGH release with modest burst release was observed for at least 6 weeks as studied by Bicinchoninic Acid (BCA) protein assay. Bioactivity of the released rhGH was determined by Nb2 cell bioassay. Our results suggest the feasibility of this system as a long-term sustained release strategy for rhGH.  This presentation will discuss the opportunties in the clinic as well as other stable formulations of active biologics from PEUs.<br/>
