α-Hydroxytropolones (αHTs) have been identified as lead scaffolds for various medicinal chemistry pursuits. One of the more promising avenues is as antivirals against herpes simplex virus -1 and -2, where an initial assessment of twenty synthetic αHTs led to the identification of <b>118</b> as the most potent lead. The following presentation will describe the synthesis of several new aryl and biaryl ketone-containing αHTs, and the evaluation of these molecules against HSV-1 and HSV-2. These molecules illustrate the important nature of the biaryl ketone appendage and provide direction in further optimizing aHTs as antivirals against herpes simplex virus.
