The use of polymer scaffolds as macromolecular therapeutic agents have been shown to be highly effective at both overcoming drug resistant infections and preventing the onset of resistance over the course of multiple treatment passages. Here, we discuss the development of a new series of cationic polycarbonates for use as macromolecular therapeutics against drug resistant diseases and infections. These polycarbonates can self-assemble into stable, well-defined nanostructures and, through preliminary <i>in vitro</i> studies, have been demonstrated to be highly effective in treatment of drug resistant infections. Mechanistic studies indicate a non-specific mode of action, making resistance onset after multiple treatment passages very difficult. Overall, this new class of cationic polycarbonates holds significant promise for dramatically improving treatment regimens for traditionally intractable diseases.<br/>
