Macrocyclic compounds play an important role in many research fields including drug discovery and development, bioorganic chemistry, and materials sciences. Sugar containing macrocycles are especially interesting compounds and they have found applications as catalysts in asymmetric synthesis, as ligands or sensors for cation or anion recognitions, and in preparation of functional supramolecular assemblies. Because of their unique structures and functions, many synthetic strategies have been developed towards the construction of novel carbohydrate based macrocycles. In this research, we utilized several different methods including copper catalyzed cycloaddition reaction, nucleophilic substitution, Glaser coupling reaction, and Ugi reaction strategies to construct novel sugar based macrocyclic scaffolds. The synthesis progress and self-assembling properties of these compounds will be discussed.
