Enzyme immobilization plays an increasingly important role in protein-protein studies, proteomics and biosensor applications. Current electron beam nanofabrication methods for immobilizing multiple enzymes are often limited by low enzyme density or decreased activity, particularly after extensive chemical modifications. Moreover, retaining protein activity after repeated sensor use is a challenging task for most devices. We report development of a novel electron beam resist material from semi-branched polyglycidols and an enzyme immobilization strategy that utilizes protein-loaded nanogels which have been directly patterned in micron dimensions to a silicon surface for enhanced signal propagation of enzyme cascade reactions. Polyglycidol crosslinking was initiated from electron beam irradiation without use of toxic initiators. As proof of concept, we developed a biochip via immobilization of three enzyme-nanogels that work synergistically towards analyte detection. The effect of enzyme encapsulation resulted in higher colorimetric signal detection compared to non-encapsulated enzyme, even after repeated uses.<br/>
