Given the modern trend in medicinal chemistry – “Escape the Flatland” – saturated 3D-shaped building blocks do play an important role.&lt;span style="font-size:10.8333px"&gt; &lt;/span&gt;Compared to their aromatic 2D-shaped counterparts, the saturated analogues usually possess higher water solubility, higher activity and lower toxicity.<br/>In this work, therefore, we have rationally designed and synthesized a library of novel saturated bioisosters of benzene. Details of the synthesis and application of the obtained compounds will be discussed.
