We have designed an undergraduate laboratory curriculum using a very stable bacterial protein, small laccase (SLAC) from <i>Streptomyces coelicolor</i>, to teach protein expression, purification, and characterization. The SLAC coding sequencing were amplified by PCR and cloned into pET15b <i>E. coli</i> expression vector. The protein were expressed in BL21(DE3) by IPTG induction. We purified the protein by affinity chromatography to exhibit the protein-purification technique. After purification, the protein was characterized for its pH optima by using enzyme kinetics.
