Lipid and carbohydrate analyses are routinely performed in many research & clinical fields. There are several methods for the quantification of total lipids and carbohydrates including gravimetric, spectrophotometric, and chromatographic etc. Although these methods are accurate & reliable, they are expensive, require large equipment and trained operator; therefore are not suitable for screening of large number samples. In this work, we have developed paper-based analytical method for the determination of total lipids and carbohydrates. The paper-based methods take advantage of well-established colorimetric assays for lipids and carbohydrates. The reactions were allowed to occur in a confined hydrophilic regions created by hydrophobic boundaries on a glass microfiber membrane. Once the reactions were complete, smartphone camera was used to take images of the paper device. The signal in each reaction zone was quantified using image-processing software and was found to be linearly co-related with the concentration of target analyte. The new lipid and carbohydrate assays method are low-cost, high throughput, and easy to use alternative to traditional methods. The new assay methods require only couple of microliters of sample and reagents therefore reduce waste generation. Application of the new assay methods in biological samples will be presented.
