In recent years, our laboratory has developed the thioamide, a single O to S substitution in canonical amide bonds, as a versatile probe for fluorescence and protease studies. In this talk will outline imroved incorporation methods of thioamides into proteins and demonstrate the structural effect that thioamide substitution has on the protein structure.
