Diabetic foot ulcers and other chronic wounds can take months or longer to heal. Wound healing is a complex process with multiple stages to completion. Chronic wounds commonly have an extended period of inflammation before being able to reach the last stages of healing. A new synthetic peptide drug called alpha Connexin carboxy Terminus 1, or aCT1, promotes wound healing and decreases inflammation. Clinical trials have shown that aCT1 significantly reduces chronic wound healing time when applied topically in a gel. However, the body metabolizes the relatively small molecule in about 24 hours or less, which is inconvenient and expensive for long-term medication. Double-emulsion solvent-evaporation method has previously been used for encapsulation and controlled release of aCT1, however encapsulation efficiency and drug loading are impractically low. Here, a flash nanoprecipitation method is used with poly(lactic-co-glycolic acid) (PLGA) to encapsulate aCT1 with higher loading and encapsulation efficiency in a process scalable for potential industrial use.<br/>
